Here's how to unlock all seven alternate skills for Enforcer and Acolyte in Risk of Rain 2: Hallowed Concepts.
Enforcer has four challenges and Acolyte has three in Risk of Rain 2, and each one unlocks an alternate skill. The skills appear in the Loadout tab on the character select screen, in the slot they replace. Each challenge counts only when you complete it as that survivor.
Both survivors come from Hallowed Concepts. Enforcer is unlocked from the start, while Acolyte unlocks through the Soulbound Threshold route.
Enforcer: Burning Lead
Burning Lead asks you to shoot a Magma Worm 24 times in 12 seconds. It unlocks Shrapnel Grenade, an alternate primary. It fires an arcing grenade for 500% damage, and in the defensive stance it fires a cluster grenade for 3x300% damage.

Turn on the Artifact of Command, and pick up a few Backup Magazines plus attack speed items such as Soldier's Syringe. Each time you swap into or out of the defensive stance, the gun reloads fully. A few swaps in a row give you the shots you need.
Magma Worms spawn on Rallypoint Delta, Abyssal Depths, and Siren's Call. Rallypoint Delta is the fastest. If an Imp Overlord or Clay Dunestrider spawns as the boss instead, move to the next stage. Turning off the other DLCs keeps you from landing on a stage with no worm.
Enforcer: +Parry in Risk of Rain 2
+Parry asks you to parry a Wandering Vagrant's supernova. It unlocks Surge Protection, an alternate secondary. You take an expanded, time-limited defensive stance. When it expires, you expend all your extra health into a large explosion for up to 1800% damage.

Wandering Vagrants can spawn on the first stage of a Risk of Rain 2 run, so reset until one does. Lower its health until it uses the supernova. Stay in the defensive stance, and press your secondary again right after the attack goes off.
Enforcer: Safe and Sound and Indomitable in Hallowed Concepts
Safe and Sound asks you to block the attacks of 12 enemies during a single teleporter event. It unlocks Ripcord, an alternate utility. You launch a remote-detonated grenade, and retriggering it creates a Pulling Blast for 300% damage that taunts enemies.
Hold your shield up during the event. Playing Enforcer for long enough likely covers it.
Indomitable asks you to survive an entire Stone Titan's laser without blocking it. It unlocks Crash Point, an alternate special. You leap high into the air, then crash down for 700% damage.

Many first-stage variants in Risk of Rain 2 have a Stone Titan, so reset until one appears. Enforcer's base health and defense do most of the work, so you need no survival items. If the first laser doesn't register, wait for the second. The Artifact of Command and Repulsion Armor Plate, which gives flat damage reduction, help.
Acolyte: Bloodbath and Not Even Close
Bloodbath asks you to kill 15 enemies within one transformation into the Anima Harvester. It unlocks Soul Jar, an alternate secondary. You throw a jar that deals 150% damage and haunts enemies. Haunted enemies take 50% damage per second and release seeking wights on death.
A normal Risk of Rain 2 run usually covers it. The Artifact of Swarms doubles spawns if you want help.

Not Even Close asks you to keep an Elder Lemurian airborne for 11 seconds. It unlocks Javelin, an alternate utility. You throw a piercing glaive for 400% damage that stuns. It explodes on large enemies and terrain for 400% damage, and reactivating it dashes you for 300% damage.
Use the Artifact of Command to take a Primordial Cube from an equipment chest. Go to a stage with Elder Lemurians, such as Abyssal Depths, Sky Meadow, or Gilded Coast. Look straight up and use the cube.
Acolyte: Hit List
Hit List asks you to land a killing blow with Sacrifice on 20 different types of monsters in a single run. It unlocks Anima Echo, an alternate Litany. You invoke an Echo of Acolyte, and during the Litany your melee attacks hit an extra time. All your missing health converts into recoverable health.

The types must differ, so beetles, Lemurians, Elder Lemurians, wisps, beetle guards, and beetle queens each count once. Elite variants reportedly don't count. Use Sacrifice at least once on every enemy on the stage.
The Artifact of Swarms and Artifact of Dissonance raise your chances of seeing new types, and the Artifact of Glass boosts your damage. The challenge takes a while, so plan for a full run.
What to Do Next
Start with the first-stage challenges in Risk of Rain 2, +Parry and Indomitable, and work on Safe and Sound alongside them. Bloodbath comes through normal play. Save Hit List for last, since it takes the longest.
Once you have the Hallowed Concepts skills, equip them in the Loadout tab on the character select screen.
To unlock the alternate loadouts in Risk of Rain 2: Hallowed Concepts, you finish seven challenges: Burning Lead, +Parry, Safe and Sound, and Indomitable for Enforcer, and Bloodbath, Not Even Close, and Hit List for Acolyte.
FAQ
Q. Where do you equip the alternate skills?

A. Yes, since Enforcer and Acolyte are survivors from that expansion.

A. +Parry and Indomitable. Wandering Vagrants and Stone Titans can spawn there, so you can reset the stage until one appears.
