Here’s how to unlock the desert cave with your Skull Key in Stardew Valley.
You get the Skull Key from the chest on floor 120 of The Mines in Stardew Valley. This guide shows where the key goes in the Calico Desert and what else it unlocks back in town. Details reflect version 1.6.
Reach the Calico Desert in Stardew Valley
In Stardew Valley, the key's door is out in the desert, which means the Bus has to run first. Finish the Vault bundles at the Community Center, or buy the bus repair through the Joja Community Development Form.

Once it runs, ride from the Bus Stop to the desert. A Warp Totem: Desert or the Desert Obelisk from the Wizard also takes you there, and the totem lands you about 4 hours earlier than the bus.
Open the Skull Cavern Door
From where you arrive, head north and a little west. Skull Cavern sits in the northwest corner of the Calico Desert.
Walk up to the locked door there with the Skull Key in your wallet, and it opens. There's nothing to select in your inventory, because the key works straight from your wallet.

What's Inside Skull Cavern in Stardew Valley
Skull Cavern in Stardew Valley has no bottom, and there's no elevator. Every visit starts again at floor 1, though a ladder at each floor's starting point lets you leave.
Your odds of finding Iridium Ore rise the deeper you go. Some monsters also drop Iridium Ore and Iridium Bars.
Each in-game hour lasts about 25% longer inside, which buys you extra floors per day. Pass out at 2:00 a.m. and you lose 1,000g or 10% of your money, whichever is less.

Play Junimo Kart in Stardew Valley
The key also opens the Junimo Kart machine on the right side of Stardew Valley's Stardrop Saloon. Beat all six levels in Progress Mode, and a Junimo Kart Arcade System arrives in the mail the next day.
What to Do Next
Floors don't save between visits, so craft Staircases from 99 Stone each and place one to drop a floor. Bring food to refill your health and energy. Head to the desert early in the day to get the most time underground. Mr. Qi reportedly sends a challenge by mail the day after your first visit. With each run, push a few floors deeper than the last.
With the Bus fixed, the Skull Key opens the cave in the northwest corner of the Calico Desert, and it unlocks Junimo Kart in town too.
